China-based Zhongsheng Pharmaceutical has reported positive results from REBUILDING-2, the first Phase III trial of a GLP-1 receptor and GIP receptor dual agonist dosed every two weeks, with 641 Chinese adults with obesity or overweight randomised to bilpatide 3 mg, 6 mg or 9 mg versus placebo for 52 weeks. The 9 mg arm reduced body weight by 22.24% from baseline, or 20.46% after placebo adjustment (p<0.0001).
99.3% of patients achieved at least 5% weight loss, 82.1% achieved at least 15% and 53.8% achieved at least 20%, while waist circumference, blood pressure, triglycerides, LDL-cholesterol, uric acid, hepatic fat and visceral fat all improved dose-dependently versus placebo.
The asset's commercial edge is convenience plus potency: bilpatide's pharmacokinetic profile supports a twice-monthly injection, a differentiated dosing rhythm against weekly tirzepatide, semaglutide and retatrutide-class molecules, and in a Chinese-setting comparison the 9 mg every-two-weeks arm beat the 15 mg weekly tirzepatide arm from SURMOUNT-CN on the share of patients losing at least 5% and at least 15% of body weight.
Gastrointestinal events remained the dominant toxicity but were mostly mild, with diarrhoea, nausea, vomiting and constipation numerically lower than or comparable to tirzepatide reports, giving Zhongsheng a credible China-first metabolic franchise that could later be positioned for global licensing.
