China-based SciBrunch Therapeutics has granted MSD exclusive worldwide rights to develop, manufacture and commercialise SPR2015, a preclinical oral molecular-glue KRAS G12D (ON) inhibitor, in a deal worth up to USD 2.13 billion including a USD 400 million upfront payment, the highest upfront ever recorded for a global out-licensing of a preclinical asset.
SPR2015 shows nanomolar antiproliferative activity across KRAS G12D-mutant tumour cell lines with strong selectivity against KRAS wild-type, and demonstrated single-agent antitumour activity in multiple CDX and PDX models, targeting the most common oncogenic RAS mutation in human cancer, prevalent in pancreatic, colorectal and lung malignancies.
