Eli Lilly's triple-targeting agonist retatrutide (GLP-1R/GIPR/GCGR) has achieved its primary and key secondary endpoints in the Phase III TRIUMPH-1 trial. The 80-week study enrolled 2,339 adults with obesity or overweight and at least one weight-related comorbidity, but without diabetes. Participants receiving weekly 12 mg doses achieved an average weight reduction of 28.3%, significantly outperforming the 2.2% reduction in the placebo group. In a subset of patients followed for 104 weeks, the weight loss increased to approximately 30.3%.
The therapy also demonstrated significant improvements in cardiovascular risk factors, including waist circumference and systolic blood pressure. Adverse events were consistent with the incretin class, primarily gastrointestinal (nausea, diarrhoea, constipation). Discontinuation rates due to adverse events were 11.3% in the 12 mg group versus 4.9% for placebo. These results position retatrutide as a potential leader in the next generation of obesity pharmacotherapy, pending regulatory submissions.
