China-based Hutchison China MediTech (HutchMed) has granted GSK an exclusive licence to HMPL-A830, a first-in-class antibody-targeted conjugate (ATTC) pairing an anti-EGFR antibody with a highly selective KRAS small-molecule inhibitor payload, for all territories outside mainland China, Hong Kong, Macau, and Taiwan, in a deal carrying USD 110 million upfront and up to USD 1.295 billion in aggregate development, regulatory, and commercial milestones, plus tiered royalties on net sales.
HutchMed runs the global Phase I programme, expected to start in the second half of 2026, with lead indications in colorectal cancer, pancreatic cancer, and lung cancer, while GSK assumes all later-phase development and commercialisation in ex-Greater China and holds a right of first negotiation on one earlier ATTC candidate.
Unlike cytotoxic-payload ADCs, the ATTC format uses a targeted signalling inhibitor as the warhead, so HMPL-A830 aims to co-localise EGFR blockade and intracellular KRAS inhibition in EGFR-expressing tumours, addressing upstream feedback resistance that limits standalone KRAS inhibitors and offering a wider therapeutic index in preclinical models.
