Monte Rosa Expands Target Space for Molecular Glues

Monte Rosa Therapeutics' 3 July Science cover paper revealed how its artificial intelligence (AI) platform identified 1,600+ human proteins targetable by cereblon (CRBN)-based molecular glue degraders (MGDs), expanding beyond traditional β-hairpin G-loop motifs to include helical G-loops. The study redefined neosubstrate recognition rules by discovering atypical binding modes like VAV1, a key immunology target, potentially enabling degradation of previously undruggable proteins.

The findings validate Monte Rosa's QuEEN discovery engine, which underpins partnerships with Roche (USD 500 million upfront) and Novartis (USD 1.5 billion deal for VAV1-targeting MRT-6160). CEO Sharon Townson noted the research accelerates first-in-class MGD development for cancer and neurological diseases, building on 2024's USD 375 million Series C financing.
