Novartis' Cosentyx Fails Phase III Trial in Giant Cell Arteritis

Novartis reported on 3 July that its interleukin-17A (IL-17A) inhibitor Cosentyx (secukinumab) missed the primary endpoint in the Phase III GCAptAIN trial for giant cell arteritis (GCA), failing to demonstrate superior sustained remission rates versus placebo at 52 weeks. Cosentyx, the first approved IL-17A antibody, currently holds indications for plaque psoriasis, psoriatic arthritis and axial spondyloarthritis among others.

Despite this setback, Novartis continues investigating Cosentyx in polymyalgia rheumatica (Phase III) and maintains a broad immunology pipeline including BTK inhibitor remibrutinib, BAFF-R inhibitor ianalumab and NLRP3/IL-18 pathway modulators. GCA, a vasculitis affecting medium/large arteries, represents an area of high unmet need with limited treatment options beyond glucocorticoids.
